Abstract

Exosomes derived from human umbilical cord mesenchymal stem cells (hucMSC-Ex) have demonstrated significant potential in cellular protection

immune modulation

and tissue regeneration. Despite these promising properties

the specific mechanisms underlying hucMSC-Ex-mediated effects in ulcerative colitis and inflammatory regulation remain unclear. This investigation focuses on elucidating the therapeutic role of hucMSC-Ex in ulcerative colitis through modulation of the miR-206/A3AR signaling axis. Using the human colon epithelial cell line HCoEpiC

we established an in vitro inflammatory model through LPS induction to evaluate the reparative effects of hucMSC-Ex on intestinal inflammation. LPS stimulation resulted in elevated levels of pro-inflammatory cytokines (TNF-α

IL-6

IL-1β) and reduced expression of the anti-inflammatory cytokine IL-10

which were subsequently reversed by hucMSC-Ex treatment. Functional assays revealed that hucMSC-Ex significantly enhanced the viability of LPS-compromised intestinal epithelial cells.Molecular analysis demonstrated upregulated miR-206 expression in hucMSC-Ex-treated cells

suggesting exosome-mediated transfer of miR-206 to intestinal epithelial cells. Through dual-luciferase reporter assays

we identified specific binding sites for miR-206 within the 3'UTR region of A3AR mRNA. These findings collectively indicate that hucMSC-Ex alleviates ulcerative colitis by delivering miR-206 to target A3AR

thereby reducing intestinal inflammation and promoting tissue repair.
